摘要: Melting temperatures, Tm, were systematically studied for a set of 92 DNA duplex oligomers in variety sodium ion concentrations ranging from 69 mM to 1.02 M. The relationship between Tm and ln [Na+] was nonlinear over this range concentrations, the observed melting temperatures poorly predicted by existing algorithms. A new empirical derived UV data that employs quadratic function, which better models as concentration is varied. Statistical analysis shows improved salt correction significantly more accurate than previously suggested algorithms predicts salt-corrected with an average error only 1.6 °C when tested against independent validation measurements obtained literature.
