摘要: Abstract Objectives Development and validation of a selective, robust high-performance liquid chromatography–tandem mass spectrometric (HPLC/MS–MS) method for the quantification morphine, morphine-3-β-glucuronide, morphine-6-β-glucuronide, hydromorphone, normorphine in human serum. Design methods Drug-free serum samples spiked with were prepared by protein precipitation using methanol containing internal standards. Samples injected onto Thermo Scientific AccuCore PFP column chromatographic separation. Detection was achieved TSQ Vantage spectrometer. Assay followed new Clinical Laboratory Standards Institute (CLSI) C62-A guidelines. Results The analytical measuring range all analytes determined to be 5 1000 ng/mL. Intra- inter-assay precision three quality control levels ≤ 7.0% ≤ 13.5%, respectively. Carryover, stability, linearity, matrix effects, extraction processing efficiency comparison characteristics acceptable relative CLSI C62 Conclusion this HPLC–MS/MS demonstrated rapid assay normorphine.
