Ugene, a Newly Identified Protein That Is Commonly Overexpressed in Cancer and Binds Uracil DNA Glycosylase

摘要: Expression microarrays identified a novel transcript, designated as Ugene, whose expression is absent in normal colon and adenomas, but that commonly induced malignant cancers. These findings were validated by real-time PCR Northern blot analysis an independent panel of cancer cases. In addition, Ugene was found to be elevated many other common types, including breast, lung, uterus, ovary. Immunofluorescence V5-tagged revealed it have nuclear localization. pull-down assay, uracil DNA glycosylase 2 (UNG2), important enzyme the base excision repair (BER) pathway, partner protein binds Ugene. Coimmunoprecipitation Western confirmed binding between endogenous UNG2 proteins. Using deletion constructs, we find first 25 amino acids NH2 terminus. We suggest induction may contribute phenotype interacting with BER pathway. [Cancer Res 2008;68(15):6118–26]
