摘要: In order to investigate the influence of DNA extraction on two PCR-based HPV genotyping tests (Linear Array, Roche and INNO-LiPA Extra, Innogenetics), three different procedures were used purify from 28 cervico-vaginal samples tested previously by Hybrid Capture 2: AmpliLute Liquid Media Extraction kit (Roche), QIAamp Blood mini (QIAGEN), NucliSENS EasyMAG automated platform (bioMerieux). All HC2-positive found positive both assays, independently extract used. Type-specific concordance (i.e., identical type-specific profile in all extracts same sample) was observed 55% 75% cases testing Linear Array INNO-LiPA, respectively. Using extracted with manual methods results concordant for INNO-LiPA. When comparing obtained either those following extraction, 65% showed cases. The 80% versus extract, while 90% when QIAGEN extract. conclusion, are affected method extraction. Consequently, profiles may be using sample. use consistent protocols purification is a priority guarantee intra-assay reproducibility over time.
