A Survey of the Humoral Immune Response of Cancer Patients to a Panel of Human Tumor Antigens

摘要: Evidence is growing for both humoral and cellular immune recognition of human tumor antigens. Antibodies with specificity antigens initially recognized by cytotoxic T lymphocytes (CTLs), e.g., MAGE tyrosinase, have been detected in melanoma patient sera, CTLs NY-ESO-1, a cancer-testis (CT) antigen identified autologous antibody, recently identified. To establish screening system the response to autoimmunogenic antigens, an enzyme-linked immunosorbent assay (ELISA) was developed using recombinant MAGE-1, MAGE-3, SSX2, Melan-A, tyrosinase proteins. A survey sera from 234 cancer patients showed antibodies NY-ESO-1 19 patients, MAGE-1 3, MAGE-3 2, SSX2 1 patient. No reactivity these found 70 normal individuals. The frequency antibody 9.4% 12.5% ovarian patients. Comparison phenotype 62 stage IV that all NY-ESO-1+ had tumors, no NY-ESO-1− tumors antibody. As proportion melanomas expressing 20–40% only this would suggest high percentage develop NY-ESO-1.
