Modification of topoisomerase genes copy number in newly diagnosed childhood acute lymphoblastic leukemia.

摘要: Topoisomerase genes were analyzed at both DNA and RNA levels in 25 cases of newly diagnosed childhood acute lymphoblastic leukemia (ALL). The results molecular analysis compared to risk group classification children order identify characteristics associated with response therapy. At diagnosis, allelic imbalance topo-isomerase IIalpha (TOP2A) gene locus was found 75% informative whereas topoisomerase I IIbeta loci are altered none or only one case, respectively. By semi-quantitative Polymerase chain reaction, we a 2.5 8-fold TOP2A amplification 72% the children, which correlated overexpression every case. These show that is frequent event ALL diagnosis. Interestingly, also identified small population do not present who significantly very high classified patients showing glucocorticoid resistance. In conclusion, characterization status diagnosis provides useful complementary information for assessment.
