Increasing the sonoporation efficiency of targeted polydisperse microbubble populations using chirp excitation

摘要: The therapeutic use of microbubbles for targeted drug or gene delivery is a highly active area research. Phospholipid-encapsulated typically have polydisperse size distribution over the 1 to 10 μm range and can be functionalized molecular targeting loaded with drug-carrying liposomes. Sonoporation through generation shear stress on cell membrane by microbubble oscillations one mechanism that results in pore formation improve delivery. A oscillating at its resonant frequency would generate maximum membrane. However, because nature phospholipid microbubbles, frequencies exist single population. In this study, linear chirp excitations was compared equivalent duration acoustic pressure tone when measuring sonoporation efficiency human colorectal cancer cells. 3 7 MHz had greatest 26.9 ± 5.6%, 16.4 1.1% 1.32 3.08 chirp. 2.2- 5-MHz efficiencies 12.8 2.1% 15.6 1.1%, respectively, which were all above 4.1 3.1% from control exposure.
