摘要: An SV40-based shuttle vector, pZ189, was used to characterize the mutation specificity and explore mechanism of chromium mutagenesis in mammalian cells. We showed previously that mutagenic DNA damage is induced by treatment plasmid with chromium(VI) plus glutathione vitro. The pattern suggested can be generation reactive oxygen intermediates. To further investigate mutagenesis, we treated cultured cells containing normal pZ189 vector chromium(Vl). Mutations were Cr(Vl) a dose-dependent manner. majority base substitution mutations widely distributed across supF target gene occurred mainly at GC basepairs. Overall, spectra not significantly different from each other except for hot spot position 43, observed only plasmids Cr(Vl)-treated characteristics Cr(VI)-induced similar those H 2 O or plasmid-containing These results are consistent hypothesis induction occurs through oxidative damage.
