A comparison of plasma levels of L(+) pseudoephedrine following different formulations, and their relation to cardiovascular and subjective effects in man.

摘要: Plasma concentrations of L(+)pseudoephedrine administered in clinically used dosages were determined by gas liquid chromatography using a nitrogen sensitive detector. They measured after administration an immediate release formulation (Sudafed) given either single dose 180 mg, or three divided doses 60 and also two different sustained preparations containing mg. Ten subjects each received five treatment regimes, being ordered balanced design based on 2 sided Latin squares. Significant differences found between plasma rates urinary excretion following the preparations. Peak greatest mg preparation while more elevations concentration followed both repeated doses. Despite these significant heart rate, blood pressure, subjective ratings mental state rarely occured, reasons for this are discussed. In second study, one was to 10 at twice daily weeks, effects measured. levels reached plateau 3 days producing increased rate initially insomnia occurred but disappeared days.
