摘要: We have recently demonstrated that human dermal fibroblasts express both constitutive and inducible nitric oxide synthases (NOS) produce (NO). In this study, NOS expression NO production were compared in derived from hypertrophic scar (HSc) site-matched normal dermis. HSc are significantly reduced when with fibroblasts. Without stimulation, the produced 7.37 +/- 1.17 nmol 11.15 0.79 nmol/10(6) cells/96 h (paired t test, p < 0.01, n = 6) as determined by Griess reaction. After stimulation 200 units interferon-gamma per ml 40 microg lipopolysaccharide ml, higher NO, indicating retain capacity to (iNOS). The Ca2+-dependent (cNOS) activity cytosol of (1.43 pmol/min/g protein) was lower than (2.60 protein), citrulline assay (p 4). mRNAs for endothelial cNOS iNOS detectable RT-PCR. Flow cytometry confirmed untreated expressed less protein Because markedly inhibits cell proliferation, our results suggest after thermal injury, tissue may undergo phenotypic alteration leading expression. low levels endogenous might be responsible cellularity post-burn HSc, a characteristic feature fibrotic condition.
