摘要: The aim of the study was to detect proteomic markers usable distinguish colorectal carcinoma from colon adenoma for a better understanding molecular mechanisms in process tumourigenesis. Therefore, we microdissected tissue, epithelial tissue as well normal adjacent epithelium and determined protein profiles by SELDI-TOF MS. A multitude significantly different signals detected. For their identification biopsis were lysed subjected two-dimensional gel electrophoresis separation. Subsequently, identified nearly 100 proteins tryptic digestion, peptide fingerprint mapping database search. Calgizzarin (S100A11; S100C) correlated very with differentially expressed signal found prior profiling. Using an immunodepletion assay confirmed identity this calgizzarin. To localise calgizzarin tissues performed immunohistochemistry. further confirmation re-analysed IHC-positive IHC-negative sections on ProteinChip arrays. This work demonstrates that biomarkers cancer can be detected, assessed approach comprising tissue-microdissection, profiling immunological techniques.
