摘要: Granulocyte-macrophage CSF (GM-CSF) primes human neutrophils for increased functional responsiveness to a variety of inflammatory agonists. In the present report, we have investigated effect GM-CSF on ability platelet-activating factor (PAF) induce synthesis 5-lipoxygenase products in neutrophils. Human stimulated with PAF range 10(-5) 10(-7) M 15 min released small quantities leukotriene B4 and its omega-oxidation products, 20-OH- 20-COOH-leukotriene amounts that were detectable by enzyme immunoassay. Preincubation normal peripheral blood rGM-CSF enhanced time- dose-dependent manner. Treatment enabled their detection response lower concentrations (greater than or equal 10(-9) M). The receptor antagonist BN52021 inhibited GM-CSF-treated PAF. addition PAF-induced synthesis, also augmented intracellular calcium mobilization This observation prompted us examine two calcium-dependent events are essential arachidonic acid liberation, activation. itself, did not directly activate either these processes, however, it consistently markedly do so. These results indicate preincubation enhances stimulate increasing both availability activation observations provide additional evidence an important role modulation responses endogenous agonists through enhancement production potent cellular mediators such as leukotrienes.
