摘要: This review delves into the rapidly evolving field of deep tissue imaging at cellular resolution, reviewing popular clearing and staining methods in combination with light-sheet fluorescence microscopy (LSFM) including quantification three-dimensional visualization tools, applications perspective, particularly focus on preclinical cancer research drug development. The LSFM technique presented here allows an extremely fast optical sectioning for reconstruction centimeter-sized samples resolution. However, are required to receive transparent tissue. Application either autofluorescence, vivo labeling, endogenous or ex whole-mount immunolabeling enables situ morphological functional features unsectioned samples. powerful innovative opens up a new dimension analysis providing detailed comprehensive insights biology. It investigation normal pathological disease progression precise monitoring potential therapeutic interventions intact biological level.
