Adhesion to fibronectin regulates Hippo signaling via the FAK–Src–PI3K pathway

摘要: The Hippo pathway is involved in the regulation of contact inhibition proliferation and responses to various physical chemical stimuli. Recently, several upstream negative regulators signaling, including epidermal growth factor receptor ligands lysophosphatidic acid, have been identified. We show that fibronectin adhesion stimulation focal kinase (FAK)-Src signaling another regulator pathway. Inhibition FAK or Src MCF-10A cells plated at low cell density prevented activation Yes-associated protein (YAP) a large tumor suppressor homologue (Lats)–dependent manner. Attachment serum-starved fibronectin, but not poly-d-lysine laminin, induced YAP nuclear accumulation via FAK–Src–phosphatidylinositol 4,5-bisphosphate 3-kinase (PI3K) Attenuation FAK, Src, PI3K, PDK1 activity blocked stimulated by fibronectin. This can, least part, explain effects spreading on localization represents Lats-dependent component response adhesion.
