摘要: The expression of the glial fibrillary acidic protein (GFAP), a component astroglial intermediate filaments, is regulated under developmental and pathological conditions. In order to characterize DNA sequences involved in such regulations, we produced transgenic mice bearing 2 kb 5' flanking region murine GFAP gene linked Escherichia coli beta-galactosidase (beta-gal) reporter gene. Seven lines were obtained. We observed that regulatory elements present transgene GFAP-nls-LacZ direct an neural non-neural tissue target vivo unexpected subpopulation astrocyte. developing brain, beta-gal activity appeared simultaneously same region, on embryonic day 18 (E18), suggesting promoter contains responsible for perinatal vimentin/GFAP switch. addition, demonstrated sequence used possess which are activated after surgical injury, thus permitting study some aspects reactive gliosis these mice.
