摘要: The proto-oncogene c-kit, encoding a receptor-type tyrosine kinase, is allelic with the W locus of mouse. stromal cell line OP9, capable supporting long-term hematopoiesis, was newly established from newborn B6C3F1-op/op mouse calvaria. When bone marrow cells WBB6F1-W/Wv mice were cocultured OP9 in liquid medium, hematopoiesis declined to level one-thousandth that cocultures WBB6F1-+/+ and by day 21. In contrast, when W/Wv semisolid at least 61% number colonies detected until end our observation period 42 days compared control cocultures, although formed hematopoietic stem significantly smaller than those normal cells. After 24-hour incubation cells, fewer ones adhered Adhesion inhibited addition an antagonists anti-c-kit monoclonal antibody, ACK2. These results demonstrate c-kit receptor plays important role not only proliferative response but also their adhesion
