Development of Osteoclasts From Embryonic Stem Cells Through a Pathway That Is c-fms but not c-kit Dependent

摘要: Osteoclasts are hematopoietic cells essential for bone resorption. To study the derivation of these interesting cells, we developed a stepwise culture system where stromal promote embryonic stem (ES) to differentiate into mature osteoclasts. Three phases this differentiation process include (1) induction hematopoiesis, along with generation osteoclast precursors, (2) expansion and (3) terminal osteoclasts in presence 1α,25-dihydroxyvitamine D3 . Although transition ES lineage was not blocked by an antibody c- fms , later were dependent on signaling through transmembrane receptor as indicated finding that anti–c- treatment second third reduced number produced 75% more than 99%, respectively. Blockade another tyrosine kinase–type receptor, kit did affect any stages osteoclastogenesis, although other hemopoietic lineages less 10% untreated. When small numbers directly cultured under conditions differentiation, tartrate-resistant acid phosphatase-positive multinucleated observed at edge but inside colonies. This suggests some types cell-cell interactions may inhibit development The here provides important tool biology.
