摘要: The advent of single-molecule biology has allowed unprecedented insight into the dynamic behavior biological macromolecules and their complexes. Unexpected properties, masked by asynchronous myriads molecules in bulk experiments, can be revealed; equally importantly, individual members a molecular population often exhibit distinct features properties. Finally, approaches allow us to study under applied tension or torsion: understanding mechanical properties these helps understand how they function cell. aim this chapter is summarize critically evaluate single DNA chromatin fibers. use high-resolution imaging capabilities atomic force microscopy been covered, together with manipulating techniques such as optical fibers, magnetic tweezers, flow fields. We have learned lot about it responds forces. It also clear that even though fibers just begun, are expected provide wealth information concerning way its structure changes during processes like transcription replication.
