Specific labelling of the protein and lipid on the extracellular surface of purple membrane.

摘要: The ferritin/avidin/biotin labelling procedure of Heitzmann & Richards (1974) has been applied to purple membrane. Two biotin reagents differing specificity have used and the then visualised in electron microscope using avidin/ferritin conjugate. With N-hydroxy succinimide ester only protein molecules are labelled; with hydrazide, periodate-oxidised membranes, lipid for both these reagents, distribution on each side membrane correlated previous structural studies diffraction freeze-fracture microscopy. The above experiments enable three deductions be made. 1. (1) Biotin protein, almost certainly at an accessible lysine residue, occurs extracellular surface membrane. 2.
