Gene expression profiling reveals molecularly and clinically distinct subtypes of glioblastoma multiforme

摘要: Glioblastoma multiforme (GBM) is the most common form of malignant glioma, characterized by genetic instability, intratumoral histopathological variability, and unpredictable clinical behavior. We investigated global gene expression in surgical samples brain tumors. Gene profiling revealed large differences between normal tumor tissues GBMs lower-grade oligodendroglial Extensive were found among GBMs, particularly genes involved angiogenesis, immune cell infiltration, extracellular matrix remodeling. that patterns paired specimens from same GBM invariably more closely related to each other than any tumor, even when had strikingly divergent histologies. Survival analyses a set ≈70 highly expressed rapidly progressing tumors stratified into two groups differed >4-fold median duration survival. further one group, FABP7, confirmed its association with survival unrelated cohorts totaling 105 patients. Expression FABP7 enhanced motility glioma-derived cells vitro. Our thus identify validate prognostic marker both biologic significance provide series putative markers for additional evaluation.
