Characterization of microglia/macrophages in gliomas developed in S-100β-v-erbBtransgenic rats

摘要: Glioma-infiltrating microglia/macrophages are referred to as tumor-associated macrophages (TAMs). Transgenic (TG) rats expressing v-erbB, which is a viral form of the epidermal growth factor receptor, under transcriptional regulation by S100-β promoter, develop brain tumors. This study was designed clarify pathological characteristics TAMs in these experimental We carried out immunohistochemical and morphometrical analyses tumors (5 malignant glioma, 4 anaplastic oligodendroglioma, astrocytoma) that developed TG rats. with ionized calcium-binding adaptor molecule 1 (Iba1) positivity morphology activated, non-phagocytic microglia increased within around gliomas astrocytomas. The Iba1-positive tumor core were significantly more activated than non-neoplastic tissue intraparenchymal oligodendrogliomas. Iba1 expression showed significant positive correlation Ki-67 all gliomas. Most no or little against CD68, CD163 CD204, although CD204-positive observed necrosis well proliferating vascular wall. In conclusion, S-100β-v-erbB may serve useful animal model for further analysis terms cell proliferation, microvascular proliferation phagocytosis, tool therapeutic use gliomas, it should be noted polarization toward M2 phenotype remains unclear.
