Enforced Expression of GATA-3 During T Cell Development Inhibits Maturation of CD8 Single-Positive Cells and Induces Thymic Lymphoma in Transgenic Mice

摘要: The zinc finger transcription factor GATA-3 is of critical importance for early T cell development and commitment Th2 cells. To study the role in development, we analyzed modified expression vivo. In mice carrying a targeted insertion lacZ reporter on one allele, found that CD4 + CD8 double-positive thymocytes correlated with onset positive selection events, i.e., TCRαβ up-regulation CD69 expression. LacZ remained high (∼80% cells) during maturation single-positive (SP) cells thymus, but developing SP fraction -expressing decreased to driven by CD2 locus control region, which provides throughout development. two independent CD2-GATA3 -transgenic lines, ∼50% developed thymic lymphoblastoid tumors were +/low mostly CD3 . tumor-free mice, total numbers thymus within normal ranges, their was hampered, as indicated increased apoptosis selective deficiency mature low HSA spleen lymph nodes, significantly reduced. These findings indicate supports lineage inhibits thymus.
