Targeting PPM1D by lentivirus-mediated RNA interference inhibits the tumorigenicity of bladder cancer cells

摘要: Protein phosphatase magnesium/manganese-dependent 1D (PPM1D) is a p53-induced that functions as negative regulator of stress response pathways and has oncogenic properties. However, the functional role PPM1D in bladder cancer (BC) remains largely unknown. In present study, lentivirus vectors carrying small hairpin RNA (shRNA) targeting were used to explore effects knockdown on BC cell proliferation tumorigenesis. shRNA-mediated significantly inhibited growth colony forming ability lines 5637 T24. Flow cytometric analysis showed silencing increased proportion cells G0/G1 phase. Downregulation also tumorigenicity nude mice. The results study suggest plays potentially important tumorigenicity, lentivirus-mediated delivery shRNA against might be promising therapeutic strategy for treatment BC.
