摘要: A high-sensitivity assay has been developed for the detection of human topoisomerase I with single molecule resolution. The method uses magnetic sepharose beads to concentrate rolling circle products, produced by amplification DNA molecules circularized and detectable a confocal microscope as discrete dots, once reacted fluorescent probes. Each dot, corresponding cleavage–religation event mediated enzyme, can be counted due its high signal/noise ratio, allowing 0.3 pM enzyme representing valid detect activity in highly diluted samples.
