摘要: Abstract Cytoplasmic 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) synthase and microsomal HMG-CoA reductase are sequential enzymes in the cholesterol biosynthetic pathway; both negatively regulated by cholesterol. In this paper, we report isolation of overlapping bacteriophage lambda clones that encompass gene for hamster cytoplasmic synthase. The spans 20 kilobases contains 11 exons 10 introns. Under conditions high-level expression cultured cells liver, approximately 50% mRNAs contain two introns region corresponding to 5' untranslated region. remaining only one intron as a result direct splicing exon 1 3. optional (exon 2) 26-nucleotide sequence is homologous mRNA reductase. Approximately 350 base pairs upstream transcription initiation site, strongly 72-base pair enhancer SV40 virus. 125 nucleotides downstream region, CCGCCC, or its inverse complement GGGCGG, repeated four times. Multiple copies present promoter flanking several other cellular housekeeping genes. availability cloned genes consecutive synthetic pathway should allow elucidation mechanism coordinate expression.
