Effects of hyaluronic acid conjugation on anti-TNF-α inhibition of inflammation in burns.

摘要: Biomaterials capable of neutralizing specific cytokines could form the basis for treating a broad range conditions characterized by intense, local inflammation. Severe burns, spanning partial- to full-thickness dermis, can result in complications due acute inflammation that contributes burn progression, and early mediation may be key factor rescuing thermally injured tissue from secondary necrosis improve healing outcomes. In this work, we examined effects on progression influence inflammatory microenvironment topical application anti-tumor factor-α (anti-TNF-α) alone, mixed with hyaluronic acid (HA) or conjugated HA. We found non-conjugated anti-TNF-α decreased macrophage infiltration greater extent than HA; however, there was little effect degree IL-1β levels. A simple transport model is proposed analyze results, which predicts qualitative quantitative differences between untreated sites those treated conjugates. Our results indicate conjugation high molecular weight HA provides sustained, modulation post-injury responses compared direct administration antibodies. © 2013 Wiley Periodicals, Inc.
