Effect of Alcohol on Glucose Production and Lactate, Pyruvate and Ketone Body Metabolism by the Isolated Perfused Rat Liver

摘要: The purpose of these experiments was to determine whether the effect ethanol on glucose production could be dissociated from its cytoplasmic DPN/DPNH ratio in presence adequate substrate. Isolated rat livers were perfused with a Krebs bicarbonate Ringer buffer containing 4 per cent albumin and 10 mM alanine increasing concentrations ethanol. Lactate, pyruvate, B-hydroxybutyrate acetoacetate used reflect cellular redox state. Despite producing maximum changes lactate, pyruvate ketone body metabolism, did not inhibit production. Ethanol 20 40 produced no further change lactate but significantly inhibited by isolated liver. These results are compatible proposal that increased entry intramitochon-drial oxidation DPNH which alcohol metabolism inhibits fatty acids, acetyl CoA generation subsequently gluconeogenesis.
