摘要: UT-7 is a human megakaryoblastic leukemia cell line with absolute dependence on interleukin-3, granulocyte-macrophage colony-stimulating factor, or erythropoietin (EPO) for growth and survival. We investigated the effect of thrombopoietin (TPO), ligand receptor encoded by c-mpl proto-oncogene, proliferation differentiation its sublines. found that UT-7/GM, which subline UT-7, but neither nor UT-7/EPO, can proliferate in response to TPO. The subline, UT-7/TPO, was established from UT-7/GM culture at lower concentrations UT-7/TPO cells had morphologically mature megakaryocytic characteristics such as developed demarcation membrane cytoplasm multinucleated appearance. This also confirmed high expression platelet factor-4 glycoprotein IIb mRNA levels level DNA content. be maintained TPO alone, doubling time 24 hours log phase. In absence TPO, majority died within few days. Thus, has an survival features. detected and, lesser degree, UT-7/GM. NF- E2 p45, reported erythroid-specific transcription upregulated whereas it down-regulated erythroid UT-7/EPO. There were no significant differences GATA-1 GATA-2 among Not only EPO induced tyrosine phosphorylation JAK2 kinase STAT5-related protein. These findings indicate would useful model analyze gene regulation maturation-associated proteins study specific actions
