摘要: DNA was extracted from approximately 600-year-old human remains found at an archaeological site in the southwestern United States, and mtDNA fragments were amplified by PCR. When these sequenced directly, multiple sequences seemed to be present. From three representative individuals, of different lengths quantified short overlapping amplification products cloned. amplifications started <40 molecules, clones contained several sequences. In contrast, when they initiated a few thousand unambiguous reproducible results achieved. These show that more experimental work than is often applied necessary ensure ancient are authentic. particular, quantitation numbers amplifiable molecules useful tool determine role contaminating contemporary PCR errors DNA.
