摘要: Acquired mutations and inherited polymorphisms in the H-ras gene may modulate risk of urinary bladder cancer. In DNA isolated from cancer tissue, we screened coding sequence H-ras, including exon-intron-junctions, for exon 1 (n = 68 patients), 2 50), exons 3 4 25). at codons 12 13 (exon 1) codon 61 2), which had been described earlier were not found any tumors, but identified a frequent polymorphism nucleotide 81T-->C wobble position. The clinical impact this was investigated case-control study 312 patients with histologically verified compared 254 hospital controls; 13.5% cases only 7.1% controls homozygous 81C-variant [odds ratio (OR), 2.04; 95% confidence interval (CI), 1.15-3.61; P 0.014]. 81C genotype overrepresented, particularly patient groups poorly differentiated tumors 145, >or=G3; OR, 2.22; CI, 1.15-4.27; 0.017), muscle-invasive 107, >or=T(2); 2.65; 1.35-5.23; 0.005), flat transitional cell carcinoma 45; 3.69; 1.60-8.51; 0.002). general, 81CC occurred more frequently advanced types We conclude that individuals harboring 81C-genotype proto-oncogene are an increased
