摘要: We have identified two additional members of the neuronal nicotinic acetylcholine receptor (nAChR)-related gene family. cDNA clones for one new gene, designated alpha 5, were isolated from rat hippocampus and PC12 cell line libraries. The 5 encodes a protein 48,800 daltons (424 amino acids) which exhibits significant overall acid sequence identity with previously cloned nAChR subunits 1 (49%), 2 (55%), 3 (52%), 4 (49%). Features characteristic other alpha-subunits are present such as conserved cysteine residues at positions 127, 141, 191, 192, four strongly hydrophobic domains. A second addition to nAChR-related family, beta 4, is encoded in overlapping genomic lambda DD15 RG518A. encoding mature 53,300 (475 acids), consists 6 exons has transcription unit length approximately 18 kilobase pairs. shows considerable (43%), (64%), (44%) subunits. Northern blots showed that, along 2, transcripts both genes line, while situ hybridization experiments demonstrated expression small number nuclei central nervous system.
