Absence of genetic alteration at codon 531 of the human c-src gene in 479 advanced colorectal cancers from Japanese and Caucasian patients.

摘要: Activation of c-src, a cellular human gene homologous in sequence to the v-src Rous sarcoma virus, had been thought play an important role progression several types cancers, without having undergone any genetic changes. However, recently truncating mutations at codon 531 c-src were reported 12% advanced colon and it was also demonstrated that this change activating, transforming, tumorigenic, metastasis promoting. To investigate whether 531-specific mutation could be involved carcinogenesis colorectal cancer Japanese Caucasian populations, we examined total 479 cancers from 421 patients (46 them with liver or lung metastases) 58 (11 metastases). Using PCR-RFLP assay additional single-strand conformation polymorphism analysis, detected no alteration cancers. Our results suggest mutational activation is unlikely significant malignant among most patients.
