Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease that generally refractory to standard chemotherapeutic agents; therefore, improvements in anticancer therapies are warranted. Poor drug delivery neoplastic cells due an extensive stromal reaction one of the major determinant therapeutic resistance PDAC. Electroporation (EP) can be used vivo increase local uptake agents by cancer (electrochemotherapy, ECT), thus leading enhanced tumour response rate. In present study, we evaluated effects reversible electroporation normal pancreas using experimental model rabbit. parallel, also tested effect increasing sensitivity pancreatic cell lines. . Materials and methods. The application ESOPE pulse protocol (1000 V/cm, 8 pulses, 100 µs, 5 KHz) rabbit was New Zealand White Rabbits short long-term toxicity assessed. PANC1 MiaPaCa2 lines were for vitro electrochemotherapy experiments. Cell permeabilization levels determined flow cytometry whereas viability pulsed cisplatin bleomycin measured trypan-blue exclusion method. Results. healthy rabbit, neither systemic nor toxic EP procedure observed, this demonstrating safety optimized electric parameters treatment established ECT where with respect without ECT.
