Abscisic acid and sucrose increase the protein content in date palm somatic embryos, causing changes in 2-DE profile

摘要: Various supplements (abscisic acid (ABA) or sucrose) were added to the initial embryo culture medium (M3) with aim of improving vigour vitroplants deriving from date palm somatic embryogenesis. ABA (20 and 40 microM) sucrose (90 g/l) applied for 4 2 weeks respectively increased thickness, no apparent difference in length. (5-40 proliferation rate. Somatic embryos maintained modified M3 (M3 supplemented an concentration) contained a higher amount protein than those (no ABA, 30 g/l sucrose), 1.5-1.7-fold increase depending on compound concentration assayed. The 1-D 2-DE profiles showed qualitative quantitative differences between cultured (control) M3. Statistical analysis spot intensity was performed by principal component analysis, yielding two accurate groups samples determining most discriminating spots. Samples also clustered using Euclidean distance average linkage algorithm. Thirty-four variable spots identified mass spectrometry analysis. Identified proteins classified into following functional categories: energy metabolism (five proteins); translation, folding degradation (9); redox maintenance (5); cytoskeleton (3); storage (2); assigned function as (10). While "up-regulation" stress-related "down-regulation" observed matured (legumin) "up-regulated" sucrose.
