Capillary electrophoresis reveals polyamine metabolism modulation in Leishmania (Leishmania) amazonensis wild-type and arginase-knockout mutants under arginine starvation

摘要: l-Arginine is an essential amino acid in Leishmania (Leishmania) amazonensis metabolism. A key enzyme for parasite l-arginine metabolism arginase (ARG) that uses arginine to produce urea and ornithine, a precursor of polyamine pathway guaranteeing replication both insect mammal hosts. There alternative ornithine via l-proline glutamate, but this mechanism not described Leishmania. In the host, two enzymes can use as substrate, host ARG induced nitric oxide synthase produces oxide. The competition between favor success infection or its control. Here, we established metabolomics profile wild type (WT) L. (L.) amazonensis, submitted starvation, compared ARG-knockout mutant (arg- ). Our results indicated starvation induces decrease arginine, putrescine, could detect significative level changes spermidine, spermine, agmatine. However, absence on arg- increase citrulline levels, decreased levels putrescine. Similarly WT arginine-starved parasites, parasites presented lower proline when ones. This be indicative surpass substrate absence.
