摘要: Aim To purify and characterize antimicrobial peptides derived from the acid extract of Gallus gallus blood cells. Methods Results Two polypeptides (i.e. CHb-1 CHb-2) with antibacterial activity were detected in acidic cells chicken (G. gallus). The isolated that possessed a potent purified using two-step chromatography procedure involved solid-phase extraction total protein/peptide followed by thin fractionation reversed-phase high performance liquid (RP-HPLC). molecular masses similar 4824·4 4825·2 Da, which have been measured matrix-assisted laser desorption/ionization mass spectrometry (MALDI TOF MS). Their amino sequences determined Edman degradation showed fully identical to two fragments G. gallus α-haemoglobin localized into different subunits (A D respectively). active micromolar concentrations against Gram-negative Escherichia coli K12 TG1. Using 1-N-phenylnaphthylamine, FITC-dextran labelled probes live/dead staining allowed show hemocidin mode action estimate pore size. Conclusion In this study, for first time, (G. gallus) has investigated as donor homologous native peptide possess in vitro. These are membrane-active their mechanism E. coli involves toroidal formation. Significance Impact Study The obtained results expand perception role haemoglobin living system, describing it source multifunction substances. Additionally, data presented paper may contribute development new, cost-effective, agents.
