摘要: To optimize tissue culture conditions for genetic transformation of Zoysia matrella (L.) Merr., we investigated the effects different plant growth regulators and medium supplements on regeneration using stolon explants excised from mature grown in a green house. Plant frequency was 33.3% when tissues with node were cultured supplemented 0.5 mg/L 2,4-D 1 kinetin. Comparing basal media tested, MS showed higher performance than N6 or SH medium. Addition 5 AgNO 3 10 cysteine improved up to 40%. Among carbon sources, 3% sucrose found show best frequency. This rapid efficient system would be useful experiments manilagrass without intervening callus-mediated regeneration.
