摘要: Recent work has shown that R-loops exist at mitotic centromeres, but the function of these is not well understood. Here, we report arise in distinct locations from those formed during interphase. They accumulate on chromosome arms prophase, where they are quickly resolved and continue to be produced repetitive sequences including centromeres a stall. Aurora B kinase activity required resolve prophase promote localization Chromosome Passenger Complex (CPC) inner centromere. CPC purified chromosomes interacts with thirty-two proteins involved R-loop biology. One these, RNA regulator RBMX, controls vivo. Perturbations homeostasis or RBMX cause defects maintenance centromeric cohesion due mislocalization CPC. We conclude generated by processes DNA sequences, play important roles fidelity, have identified set regulators will enable future studies R-loops.
