The Potential Roles of Long Noncoding RNAs (lncRNA) in Glioblastoma Development.

摘要: Long non-coding RNA (lncRNA) may contribute to the initiation and progression of tumor. In this study, we first systematically compared lncRNA mRNA expression between GBM paired normal brain tissues using microarray data. We found 27 82 significantly up-regulated in GBM, as well 198 285 down-regulated GBM. identified 138 co-expressed lncRNA-mRNA pairs from these differentially expressed genes. Subsequent pathway analysis lncRNA-paired genes indicated that EphrinB-EPHB, p75-mediated signaling, TNF alpha/NF-κB, ErbB2/ErbB3 signaling pathways might be altered Specifically, RAMP2-AS1 had significant decrease showed co-expressional relationship with NOTCH3, an important tumor promoter many neoplastic diseases. Our follow up experiment (1) overexpression reduced cell proliferation vitro also xenograft tumors vivo; (2) NOTCH3 co-expression rescued inhibitory action glioblastoma cells; (3) pull-down assay revealed a direct interaction DHC10, which consequently inhibit, hypothesize, its downstream molecule HES1 Taken together, our data profile tissue was altered; play suppressive role through indirect inhibition NOTCH3. results provided some insights into understanding key roles co-regulation human mechanisms responsible for pathogenesis.
