摘要: One of the critical variables that influences efficacy clinical neural transplantation for Parkinson's disease (PD) is optimal graft placement. The current paradigm focuses on ectopic placement fetal grafts in striatum (ST) fails to reconstruct basal ganglia circuitry or normalize neuronal activity important structures, such as substantia nigra (SN) and subthalamic nucleus (STN). aim this study was investigate a multitarget strategy PD by assessing whether simultaneous dopaminergic transplants ST, SN, STN induce functional recovery hemiparkinsonian rats. Forty-six female Wistar rats with unilateral 6-hydroxydopamine lesions nigrostriatal pathway were randomly divided into eight groups received only injections 900,000 embryonic rat ventral mesencephalic cells (1) (2) (3) STN, (4) ST (5) (6) (7) SN STN. number transplanted equally among grafting sites. Animals two 450,000 each structure, animals three 300,000 per structure. Recovery assessed amphetamine-induced rotations stepping tests. Graft survival using tyrosine hydroxylase immunohistochemistry. At 8 weeks after transplantation, induced significant improvement rotational behavior test scores. Intrastriatal associated asymmetry, whereas improved forelimb function These results suggest restoration multiple targets, promotes more complete complex sensorimotor behaviors. A transplant aimed at optimizing reinnervation may be crucial improving outcomes patients.
