摘要: Circadian rhythms are essential to health. Their disruption is associated with metabolic diseases in experimental animals and man. Local represent an output of tissue-based circadian clocks. Attempts define how local metabolism temporally coordinated have focused on gene expression by defining extensive divergent "circadian transcriptomes" involving 5%-10% genes assayed. These analyses inevitably incomplete, not least because coordination depends ultimately upon temporal regulation proteins. We therefore conducted a systematic analysis mammalian proteome." Our revealed that up 20% soluble proteins assayed mouse liver subject control. Many these novel cluster into discrete phase groups so the liver's enzymatic profile contrasts dramatically between day night. Unexpectedly, almost half cycling lack corresponding transcript, as determined quantitative PCR, microarray, or both revealing for first time extent posttranscriptional mechanisms control points. The proteome includes rate-limiting factors vital pathways, including urea formation sugar metabolism. findings provide new perspective contribution programming hepatic physiology.
