Direct Identification of Insulator Components by Insertional Chromatin Immunoprecipitation

摘要: Comprehensive understanding of mechanisms epigenetic regulation requires identification molecules bound to genomic regions interest in vivo. However, non-biased methods identify specific loci vivo are limited. Here, we applied insertional chromatin immunoprecipitation (iChIP) direct components insulator complexes, which function as boundaries domain. We found that the chicken β-globin HS4 (cHS4) complex contains an RNA helicase protein, p68/DDX5; species, steroid receptor activator 1; and a nuclear matrix Matrin-3, Binding p68 Matrin-3 cHS4 core sequence was mediated by CCCTC-binding factor (CTCF). Thus, our results showed it is feasible directly proteins region using iChIP.
