Identification of telomere-associated molecules by engineered DNA-binding molecule-mediated chromatin immunoprecipitation (enChIP)

摘要: Biochemical analysis of molecular interactions in specific genomic regions requires their isolation while retaining vivo. Here, we report telomeres by engineered DNA-binding molecule-mediated chromatin immunoprecipitation (enChIP) using a transcription activator-like (TAL) protein recognizing telomere repeats. Telomeres recognized the tagged TAL were immunoprecipitated with an antibody against tag and subjected to identification telomere-binding molecules. enChIP-mass spectrometry (enChIP-MS) targeting identified known novel proteins. The data have been deposited ProteomeXchange identifier PXD000461. In addition, showed that RNA associated could be isolated enChIP. Identified molecules may play important roles biology. enChIP proteins would useful tool for biochemical interest.
