摘要: A transient assay is described that should allow evaluation of the role host genes in disease response by enhancing or disrupting expression those specific cells and looking for effects on development. The also has potential assessing utility non-host resistance to transgenic plants. Particle bombardment with a helium discharge particle gun was utilized transiently express epidermal coleoptiles barley (Hordeum vulgare). An anthocyanin reporter gene construct provided means identifying were expressing introduced DNA. Optimal rates achieved two days following 1800 psi pressure, 1.0 μm diameter gold particles, coleoptile pre- post-treatment 0.30--0.35 m mannitol/sorbitol. Under optimal conditions, at least 35 expressed per bombardment. Transiently inoculated fungal pathogen, Erysiphe graminis f. sp. hordei, development observed. Neither procedures, presence nearby dead cells, nor accumulation within living affected cells. Therefore, incorporation disease-related onto same plasmid as will suppression. Since possible great range different plant tissues, methodology exhibit wide applicability evaluating diverse plant-pathogen interactions, well involved many other biological processes
