Caractérisation moléculaire de tumeurs pulmonaires radon-induites chez le rat

摘要: Le radon est un carcinogene pulmonaire avere chez l'homme a fortes doses mais l'existence d'un risque de cancer faibles n'est pas etablie car les approches epidemiologiques se heurtent, ce niveau doses, nombreux facteurs confondants. Ces donnees soulignent la necessite disposer modele d'etude des tumeurs pulmonaires l'animal. but travail these caracteriser, sur le plan moleculaire, une serie radon-induites rat qui comprend adenocarcinomes (AC), carcinomes epidermoides (Sq) et adenosquameux (ASq) formes composantes cellulaires AC Sq. Une analyse cytogenetique globale ces par technique CGH permis definir desequilibres recurrents regions chromosomiques, lesquelles ont ete cibles genes candidats potentiellement impliques dans developpement tumeurs, comme p16Ink4a, p19Arf, Rb1, K-Ras ou c-Myc. plus precise voies p16Ink4a/Cdk4/Rb1 p19Arf/Mdm2/Tp53 menee mettre en evidence inactivation frequente voie perte d'expression proteine indiquant que cette joue role majeur pulmonaires. Enfin, transcriptomique comparative trois types induites revele complexes ASq profil genique reflete leur nature composite qu'elles presentent egalement specificites. L'ensemble mis jour caracteristiques moleculaires communes aux murines humaines encouragent poursuivre l'etude specificites radio-induites pour aider meilleure caracterisation l'homme.
