摘要: We had developed Optical Scatter Imaging (OSI) as a method which combines light scattering spectroscopy with microscopic imaging to probe local particle size in situ. Using variable diameter iris Fourier spatial filter, the technique consisted of collecting images that encoded intensity ratio wide-to-narrow angle scatter at each pixel full field view. In this paper, we replace filter digital micromirror device (DMD) extend our assessment morphology characterization shape and orientation. describe setup detail demonstrate how eliminate aberrations associated placement DMD conjugate plane system. bacteria polystyrene spheres, show system can be used assess aspect even when imaged low resolution. also feasibility detecting alterations organelle situ within living cells. This improved OSI could further automate morphological quantification sorting non-spherical particles
