Cross-linking and other structural proteomics techniques: how chemistry is enabling mass spectrometry applications in structural biology

摘要: The biological function of proteins is heavily influenced by their structures and organization into assemblies such as protein complexes regulatory networks. Mass spectrometry (MS) has been a key enabling technology for high-throughput comprehensive identification quantification on proteome-wide scale. Besides these essential contributions, MS can also be used to study higher-order biomacromolecules in variety ways. In one approach, intact or may directly probed the mass spectrometer. Alternatively, various forms solution-phase chemistry are introduce modifications localizing analysis at peptide level derive structural information. Here, I will put spotlight central role spectrometry-based methods that bridge proteomics biology, with particular emphasis chemical cross-linking complexes.
