摘要: The tissue distribution of apolipoprotein (apo) E synthesis in the cynomolgus monkey, Macaca fascicularis, was determined via short-term organ culture with radiolabeled amino acid. Tissue extracts were reacted antiserum to apo-E, and immunoprecipitates analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Newly synthesized apo-E detected liver, adrenal, testis, lung, spleen, mesenteric lymph node, kidney. Peripheral hepatic showed same electrophoretic mobility. High resolution two-dimensional analysis that newly exists two major isoforms each examined. Comparison isoform patterns mixing experiments have identical charge properties These data indicate numerous peripheral tissues synthesize is indistinguishable from liver criteria tested. Measurements relative synthetic capacities illustrate a moderately abundant protein product variety although quantitative differences occur. Apo-E mRNA monkey human Hep G2 cells co-migrated an mobility corresponding approximately 1200 nucleotides. brain, thymus, kidney, spleen size. Primer extension yielded cDNA representing complete copying 5' untranslated region mRNA. A size produced thymus. suggest transcription gene initiated at or near site
