Development of a new multi-analyte assay for the simultaneous detection of opioids in serum and other body fluids using liquid chromatography–tandem mass spectrometry

摘要: Abstract A liquid chromatography–tandem mass spectrometry method using electrospray ionization in positive mode was developed for the simultaneous detection of multiple opioid-type drugs plasma. The presented assay allows quantitative determination alfentanil, buprenorphine, codeine, desomorphine, dextromethorphan, dextrorphan, dihydrocodeine, dihydromorphine, ethylmorphine, fentanyl, hydrocodone, hydromorphone, methadone, morphine, naloxone, naltrexone, oxycodone, oxymorphone, pentazocine, pethidine, pholcodine, piritramide, remifentanil, sufentanil, and tramadol as well metabolites 6-monoacetylmorphine, bisnortilidine, morphine-3-glucuronide, morphine-6-glucuronide, naltrexol, norbuprenorphine, norfentanyl, norpethidine, nortilidine, O -desmethyltramadol. Serum blood samples were purified by solid-phase extraction. analytes separated on a phenyl–hexyl (100 mm) column formic acid/acetonitrile gradient elution an UPLC 1290 Infinity coupled with 6490 Triple Quadrupole spectrometer. limits ranged from 0.02 to 0.6 ng/mL lower quantification 0.1 2.0 ng/mL. calibration curves linear between Calibration Levels 1–6 all 35 substances. Recovery rates 51 88% compounds except morphine-3-glucuronide. matrix effect 86% ethylmorphine 105% desomorphine. Using validation procedure proposed German Society Toxicological Forensic Chemistry, acceptable precision accuracy data almost obtained. successfully applied 206 authentic serum provided palliative intensive care units University Medical Center police authorities. Furthermore, suspected fatal intoxication is demonstrated analysis sufentanil post mortem body fluids tissues.
