摘要: In a normal mature neuron, microtubule associated protein tau promotes the assembly of tubulin into microtubules and maintains structure microtubules. Alzheimer disease brain, is abnormally hyperphosphorylated major subunit paired helical filaments (PHF). present study, biological activity in PHF effect dephosphorylation on this were examined. isolated from brains untreated or alkaline phosphatase-treated was extracted by ultrasonication buffer. Tubulin phosphocellulose chromatography three cycled bovine brain. PHF-tau did not promote whereas dephosphorylated produced robust assembly. These studies suggest (i) that functionally inactive because abnormal phosphorylation (ii) phosphorylated site(s) inactivates accessible to enzymatic vitro.
